Antibacterial activity of Spondias mombin on superinfecting bacteria in the oral environment

Resumo: Objective: To evaluate in vitro the antimicrobial action of the hydroalcoholic extract of the leaf of Spondias mombin (cajá), on Staphylococcus aureus (ATCC 25923) and Enterococcus faecalis (ATCC 29212). Methodology: The microbial strains were grown in BHI broth (Brain Heart Infusion) and incubated at 37 ° C for 18-20 hours. In petri dishes containing Mueller Hinton Agar, inoculated saline solution was introduced with each microbial growth and five standardized holes of approximately six mm in diameter were made. 50µL of the test substance (Crude Extract - EB / Substance Pure - SP until 1: 512 dilution) was introduced into the holes and incubated at 37 ° C for 24 hours. The assay was performed in duplicate on each strain. Chlorhexidine digluconate 2% and sodium hypochlorite 2.5% were used as positive controls. For data analysis, statistical parameters were used that included values: minimum, maximum, average and standard deviation. Mann-Whitney U nonparametric test was applied to compare the minimum inhibitory concentration of the substances. Results: It was observed that the chlorhexidine and the extract of the caja showed superior performance to the sodium hypochlorite 2.5% on the tested bacteria. According to the results of the Mann Whitney U test, there was a statistically significant difference only between the extract of caja and sodium hypochlorite 2.5% (p <0.05). Conclusions: It is concluded that the extract of the leaf of S. mombin showed inhibitory activity on planktonic growth of S. aureus and E. faecalis, stimulating research for the treatment of persistent endodontic infections through bioactive products.

Metodología: Las cepas microbianas se cultivaron en caldo BHI (infusión de cerebro y corazón) y se incubaron a 37 ° C durante 18-20 horas. En placas de Petri que contenían Agar Mueller Hinton, se introdujo solución salina inoculada con cada crecimiento microbiano y se realizaron cinco orificios estandarizados de aproximadamente seis mm de diámetro. Se introdujeron 50 µl de la sustancia de ensayo (extracto crudo - EB / sustancia pura - SP hasta una dilución 1: 512) en los orificios y se incubaron a 37ºC durante 24 horas. El ensayo se realizó por duplicado en cada cepa. Como controles positivos se utilizaron digluconato de clorhexidina al 2% e hipoclorito de sodio al 2,5%. Para el análisis de los datos se utilizaron parámetros estadísticos que incluyeron valores: mínimo, máximo, promedio y desviación estándar. Se aplicó la prueba no paramétrica U de Mann-Whitney para comparar la concentración mínima inhibitoria de las sustancias. Resultados: Se observó que la clorhexidina y el extracto de la caja mostraron un desempeño superior al hipoclorito de sodio al 2.5% sobre las bacterias probadas. De acuerdo con los resultados de la prueba U de Mann Whitney, hubo una diferencia estadísticamente significativa solo entre el extracto de caja y el hipoclorito de sodio al 2.5% (p <0.05). Conclusiones: Se concluye que el extracto de hoja de S. mombin mostró actividad inhibidora sobre el crecimiento planctónico de S. aureus y E. faecalis, estimulando la investigación para el tratamiento de infecciones endodónticas persistentes a través de productos bioactivos.Objetivo: Avaliar in vitro a ação antimicrobiana do extrato hidroalcóolico da folha de Spondias mombin (cajá), sobre Staphylococcus aureus (ATCC 25923) e Enterococcus faecalis (ATCC 29212). Metodologia: As linhagens microbianas foram cultivadas em caldo BHI (Brain Heart Infusion) e incubadas a 37 °C por 18-20 horas. Em placas de petri contendo Agar Mueller Hinton foi introduzida solução salina inoculada com cada crescimento microbiano e confeccionados cinco orifícios padronizados de aproximadamente seis mm de diâmetro. Foram introduzidos 50µL da substância teste (Extrato Bruto- EB/Substância Pura - SP até a diluição 1:512) nos orifícios e incubadas a 37°C por 24 horas. O ensaio foi realizado em duplicata sobre cada linhagem. Utilizou-se como controles positivos o digluconato de clorexidina a 2% e o hipoclorito de sódio 2,5%. Para análise dos dados utilizou-se parâmetros estatísticos que incluíram valores: mínimo, máximo, média e desvio padrão. Aplicou-se o teste não paramétrico U de Mann-Whitney na comparação da concentração inibitória mínima das substâncias. Resultados: Observou-se que a clorexidina e o extrato do cajá mostraram desempenho superior ao hipoclorito de sódio 2,5% sobre as bactérias testadas. Conforme resultados do teste U de Mann Whitney observou-se estatisticamente significante apenas entre o extrato do cajá e o hipoclorito de sódio 2,5% (p< 0,05).
