Resumo: | Photodynamic therapy (PDT) is a group of physical, chemical and biological procedures, which occurs after the use of a photosensitizer (PS) activated by light of a specific wavelength (Laser or Led), to destroy the target cell and / or micro-organisms. The aim of this study was to evaluate the cytotoxicity of PDT and the irrigating solutions. L-929 mouse lineage fibroblasts cells were used in this study. The cultures were maintained under standard cell culture conditions (37 ºC and 5% CO2). The solutions were distributed in groups as follow: G1 - Culture medium DMEM (control); G2 - Sodium chloride 0.9%; G3 - 2.5% NaOCl; G4 - 5% NaOCl; G5 - TFD (PS curcumin 1000 mg/L + blue Led λ 480 nm, fluency of 75 J/cm², for 4 minutes); G6 - TFD (PS curcumin 750 mg/L + blue Led λ 480 nm, fluency of 75 J/cm², for 4 minutes). After 6h, 24h and 48h, viable cells were stained with formazan dye MTT, at 37 ºC, 5% CO2 for 4h. The results were statistically analyzed by ANOVA with Bonferroni correction (p<0.05). The control group showed statistically significant difference (p<0.05) with all tested solutions, in all tested periods. There was statistically significant difference about the group of sodium chloride 0.9% with NaOCl 5% and PDT curcumin (1000 mg/L and 750 mg/L) in the period of 6h (p<0.05); with NaOCl (2.5% and 5%) and PDT curcumin (1000 mg/L and 750 mg/L) in the period of 24h (p<0.05); and with PDT curcumin (1000 mg/L) 48h (p<0.05). The NaOCl 2.5% showed less cytotoxic than NaOCl 5% and curcumin 1000 mg/L and 750 mg/L in all experimental periods. The NaOCl 5%, PDT curcumin 1000 mg/L and 750 mg/L were the most cytotoxics solutions (p>0.05) in all periods. There was not statistically significant difference between PDT curcumin 1000 mg/L and 750 mg/L (p>0.05). Conclude that PDT curcumin 1000 mg/L was cytotoxic in L-929 fibroblasts culture. O objetivo do presente estudo foi avaliar a citotoxicidade da TFD e das soluções irrigadoras. Foram utilizadas células de linhagem de fibroblastos de camundongos L-929. As culturas foram mantidas sob condições padrão de cultivo celular (37ºC com 5% de CO2). As soluções foram distribuídas em grupos: G1-Meio de Cultura DMEM (Controle); G2-Cloreto de Sódio 0,9%; G3-Hipoclorito de Sódio 2,5%; G4-Hipoclorito de Sódio 5%; G5-TFD (FS curcumina 1000 mg/L + Led azul λ 480 nm, fluência de 75 J/cm², por 4 minutos); G6-TFD (FS curcumina 750 mg/L + Led azul λ 480 nm, fluência de 75 J/cm², por 4 minutos). Depois de 6h, 24h e 48h, foi realizada a análise dos resultados da viabilidade celular pelo método colorimétrico MTT, por 4h a 37°C em 5% de CO2. Os resultados foram analisados estatisticamente pelo teste ANOVA com correção de BonFerroni (p<0,05). O grupo controle apresentou diferença estatística significante com todas as soluções testadas (p<0,05) em todos os períodos testados. Houve diferença estatisticamente significante do grupo de cloreto de sódio 0,9% com o hipoclorito de sódio (5%) e TFD curcumina (1000mg/L e 750 mg/L) no período de 6h (p<0,05); com o hipoclorito de sódio (2,5% e 5%) e TFD curcumina (1000mg/L e 750 mg/L) no período de 24h (p<0,05); e com TFD curcumina (1000mg/L) em 48h (p<0,05). O hipoclorito de sódio 2,5% apresentou menos tóxico que o hipoclorito de sódio 5%, curcumina 1000 mg/L e 750 mg/L em todos os períodos experimentais. O hipoclorito de sódio 5%, TFD curcumina 1000 mg/L e 750 mg/L, foram as soluções mais citotóxicas (p>0,05) em todos os tempos.
