In vitro vine propagation: production of basic plants for rootstocks

Resumo: Techniques in vitro are useful in order to generate basic material with high genetic quality andadequate sanitary standards. The present study aimed at the in vitro production of vine rootstocks to evaluate morphological parameters during micropropagation. Axillar buds of ‘Paulsen 1103’, ‘VR043-43’ and ‘Gravesac’ rootstocks were inoculated on DSD1 culture medium. The rate of responsive cultures was 65% and in in vitrocondition the leaf area and the dry weight were different among the genotypes, being the rootstock ‘Paulsen 1103’ the best one in terms of number of leaves, length of roots, leaf area and biomass yield. ‘Gravesac’ presented the best results for stem length and root number. The rate of survival in the acclimatization was 95%. The evaluatedrootstocks showed adequate morphologic features for micropropagation and acclimatization.

As técnicas de cultura in vitro apresentam um grande potencial para a produção de plantas matrizes e mudas de alta qualidade genética e sanidade comprovada. O objetivo deste trabalho foi estabelecer porta-enxertos de videira in vitro,avaliar os parâmetros morfológicos fundamentais à micropropagação e multiplicar plantas matrizes básicas. Os porta-enxertos ‘Paulsen 1103’, ‘VR043-43’ e ‘Gravesac’ foram introduzidos e multiplicados in vitro pelo método de gemas axilares em meio de cultura DSD1. A taxa de estabelecimento in vitrofoi de 65%, e nas condições de cultura in vitro o crescimento, a área foliar e o peso seco foram diferentes entre genótipos. O porta-enxerto ‘Paulsen 1103’ foi superior em número de folhas, comprimento de raízes, área foliar e produção de biomassa, e o ‘Gravesac’, em comprimento de caule e número de raízes. A taxa média de sobrevivência de plantas na aclimatização foi 95%.
