Resumo: ltrasound has been used for the clinical treatment of a number of inflammatory musculoskeletal conditions, but its efficacy has not yet been proven. The aim of the present study was to assess the effect of therapeutic ultrasound on the regeneration rat skeletal muscle following cryoinjury and determine its role in the expression of myogenic factors (MyoD and myogenin). One hundred Wistar rats were separated into a control group (n=10), sham group (n=10), cryoinjured group without treatment (n=40) and cryoinjured group treated with ultrasound (n=40). In the groups with cryoinjury, the injury was performed on the left tibialis anterior muscle. Among the treated animals, the muscles were submitted to daily one-minute applications of pulse ultrasound at a frequency of 1 MHz and intensity of 0.8 W/cm2. The animals in the cryoinjury and ultrasound groups were assessed after one, seven, 14 and 21 days. Body weight was determined in all evaluation periods. The muscles were morphologically analyzed using hematoxylin-eosin staining. The expression of myogenic factors (MyoD and myogenin) was determined using real-time PCR. The morphological and semi-quantitative analysis of edema, inflammatory process, myonecrosis and the presence of new fibers revealed that treatment with ultrasound promoted a reduction in the acute inflammatory process on Day 1 following injury, but did not affect the other phases of muscle repair. Ultrasound did not alter the expression of mRNA of MyoD or myogenin in any evaluation period. In conclusion, pulsed ultrasound did not accelerate the muscle regeneration process or affect the expression of proteins involved in myogenesis under the conditions analyzed. However, ultrasound proved effective in modulating the acute inflammatory process in rat skeletal muscle following cryoinjury.

O objetivo deste estudo foi avaliar o efeito do US terap??utico na regenera????o do m??sculo esquel??tico de rato ap??s crioles??o bem como, seu papel na express??o de RNAm de fatores miog??nicos (MyoD e miogenina). Foram utilizados 100 ratos da linhagem wistar separados em grupo controle (n=10), grupo sham (n=10), grupo criolesionado sem tratamento (n=40) e criolesionado tratado com US (n=40). Nos grupos criolesionados a les??o foi realizada no m??sculo tibial anterior do lado esquerdo. Os animais do grupo criolesionado e US foram avaliados ap??s 1, 7, 14 e 21 dias. Nos animais tratados os m??sculos foram submetidos a tratamento com US pulsado com intensidade de 1 MHz e intensidade de 0,8 W/cm2, esfera do cabe??ote de 3,5 cm, durante 1 minuto com aplica????es di??rias. O peso corp??reo dos animais foi obtidos em todos os tempos experimentais. Os m??sculos foram analisados morfologicamente pela colora????o de hematoxilina & eosina. A express??o de fatores miog??nicos (MyoD e miogenina) foi avaliada por PCR em tempo real. A an??lise morfol??gica e semi-quantitativa do edema, processo inflamat??rio, mionecrose e presen??a de novas fibras mostrou que o tratamento com US promoveu redu????o do quadro inflamat??rio agudo ap??s 1 dia de inj??ria por??m, n??o modificou as demais fases do reparo muscular. Al??m disso, o US n??o alterou a express??o de RNAm de MyoD e miogenina em todos os per??odos e grupos experimentais avaliados. Conclui-se que o US pulsado nas condi????es avaliadas n??o acelerou o processo de regenera????o muscular e n??o interferiu na express??o de fatores miog??nicos relacionado com a prolifera????o (MyoD) e diferencia????o (miogenina) das c??lulas sat??lites.
