Resumo: | The Persea americana Mill fruit has significant concentrations of condensed tannins considered beneficial for health and contributes to the control of the lipid, glycemic, blood pressure, body weight and even inhibition of free radicals. Antioxidants and a higher concentration of phenolic compounds are found in fruits and in parts such as seeds, which have values in their composition that reach more than 70% in relation to the pulp. So, considering the potential of Persea americana Mill despertou up interest in investigating different concentrations of the fruits of the seeds developed in José de Freitas Piauí region. With the main aim of this study to develop a byproduct from the association of parts, cored and seed the fruit Persea americana Mill, aiming to improve the control of risk factors of metabolic syndrome. The hydroethanolic extract of the associated parts: endocarp and seed of the Persea americana Mill fruit was obtained by the percolation method (7: 3) and its chemical composition characterized by HPLC-PDA and LC-MS. The in vitro activities, toxicity and antioxidant, were evaluated using DPPH and MTT, respectively. The effects of the ESP (core material Seed Extract and Persea americana Mill) for monitoring induced obesity was performed by Lee index, feed intake, TOTG and TTI. Evaluating CT, TG, HDL-C and LDL-C, as well as AST and ALT in mice with MS. As results, were obtained: Quinic acid; Catechin; Procyanidins B1; Cumáric Acid; Trimer of catechins and galatocatechins. When evaluating the antioxidant capacity, he found an IC50 of 300 µg / mL. The toxicity tested in vitro, showed that in the concentration of 500µg / mL there was 50% more cell viability when compared to the control. In the in vivo evaluation, the 125mg / kg group associated with the practice of physical exercise had the best results when compared to the CG (control) and DS (without treatment), respectively, in the CT (1%; 22%), HDL-C (66%; 78%) and in the AST / ALT index (34%; 78%). This group also stood out in the GL values, reducing 59% in relation to the DS, the TG levels were 3% below the ED (statin) and 30% in relation to the DS and the LDL-C reduced 3% more than the ED and more than 70% compared to DS. However, all groups treated with ESP had positive results for the control of risk factors for metabolic syndrome. With this finding, it was concluded that the ESP is a promising product to control the adverse events and risk to develop metabolic syndrome associated with physical exercise. From these data, alternatives can be made for therapeutic purposes, phytopharmaceuticals and / or phytotherapics. Producing a herbal medicine is the intention addressed in this study. Therefore, the improvement of the tests should be reason to continue the research and prove its real effectiveness. |

Os antioxidantes e uma maior concentração de compostos fenólicos são encontrados nos frutos e em suas partes como a semente, esta possui na sua composição valores que chegam a ser superiores a 70% em relação à polpa. Então, considerando o potencial de Persea americana Mill, despertou-se o interesse em investigar diferentes concentrações das sementes dos frutos desenvolvidos na região de José de Freitas no Piauí. Tendo como principal objetivo deste estudo, desenvolver um bioproduto a partir da associação das partes: endocarpo e semente do fruto Persea americana Mill, visando melhorar o controle dos fatores de risco da síndrome metabólica. O extrato hidroetanólico das partes associadas: endocarpo e semente do fruto Persea americana Mill foi obtida pelo método da percolação (7:3) e sua composição química caracterizada através do HPLC-PDA e LC-MS. As atividades in vitro, toxicidade e antioxidante, foram avaliadas por meio do DPPH e MTT, respectivamente. Os efeitos do ESP (Extrato do Endocarpo e Semente Persea americana Mill) sobre o monitoramento da obesidade induzida foi feita através do Índice de Lee, consumo de ração, TOTG e TTI. Avaliando-se o CT, TG, HDLC e LDL-C, assim como AST e ALT em camundongos com SM. Como resultados, foram obtidos: Ácido quínico; Catequina; Procianidinas B1; Ácido Cumárico; Trímero de catequinas e Galatocatequinas. Ao avaliar a capacidade antioxidante encontrou o IC50 de 300 µg/mL. A toxicidade testada in vitro, mostrou que na concentração de 500µg/mL houve 50% a mais de viabilidade celular quando comparado ao controle. Na avaliação in vivo, o grupo 125mg/kg associado à prática de exercício físico tiveram os melhores resultados quando comparado ao GC (controle) e ao DS (sem tratamento) respectivamente, no CT (1%; 22%), HDL-C (66%; 78%) e no índice AST/ALT (34%; 78%). Este grupo também se destacou nos valores da GL reduzindo 59% em relação ao DS, os níveis de TG ficaram 3% abaixo do DE (estatina) e 30% em relação ao DS e o LDL-C reduziu 3% mais que o DE e mais de 70% em relação ao DS. No entanto, todos os grupos tratados com o ESP tiveram resultados positivos para o controle dos fatores de risco da síndrome metabólica. Diante do achado, concluiu-se que, o ESP é um produto promissor para controlar os eventos adversos e de riscos que desenvolvem a síndrome metabólica associada ao exercício físico. A partir destes dados, alternativas poderão ser realizados com fins terapêuticos, produtos fitofármacos e/ou fitoterápicos. Produzir um fitoterápico é a pretensão tratada neste estudo. This group also stood out in the GL values, reducing 59% in relation to the DS, the TG levels were 3% below the ED (statin) and 30% in relation to the DS and the LDL-C reduced 3% more than the ED and more than 70% compared to DS. However, all groups treated with ESP had positive results for the control of risk factors for metabolic syndrome. With this finding, it was concluded that the ESP is a promising product to control the adverse events and risk to develop metabolic syndrome associated with physical exercise. From these data, alternatives can be made for therapeutic purposes, phytopharmaceuticals and / or phytotherapics. Producing a herbal medicine is the intention addressed in this study. Therefore, the improvement of the tests should be reason to continue the research and prove its real effectiveness. |
