Cashew gum (Anacardium occidentale L.) used as an alternative mounting medium in histological preparations

Resumo: In this work, the objective was to use the resinous exudate of a typical plant of the Brazilian Northeast, the cashew tree gum (Anacardium occidentale L.) as an alternative mounting medium in the histological technique, economically viable and environmentally healthy. Cashew gum was processed according to the adapted method from Rinaudo and Milas. The refractive index reading, and the viscosity measurement were performed using an Abbes refractometer and an MVD-20 digital viscometer, respectively. According to the authorization of the Animal Use Committee of the Federal University of Pernambuco, different organs of Wistar rats were histologically processed and the permanent preparations were mounted with cashew resin gum (MGRC). The refractive index of the MGRC was 1.343 and the viscosity of the medium without mineral oil was 1143.7 ± 267.4 mPas.s and with the mineral oil it was 939.9 ± 103.7 mPas.s. The preparations mounted with MGRC, regardless of the type of stain used, presented with clear images and morphology of the structures conserved. The MGRC proved to be a satisfactory alternative to commercial assembly means, being economically viable and generating benefits such as personal biosecurity as it is non-toxic, in addition to not harming the environment.

La goma de anacardo se procesó según el método adaptado de Rinaudo y Milas. La lectura del índice de refracción y la medición de la viscosidad se realizaron utilizando un refractómetro Abbes y un viscosímetro digital MVD-20, respectivamente. De acuerdo con la autorización del Comité de Uso Animal de la Universidad Federal de Pernambuco, se procesaron histológicamente diferentes órganos de ratas Wistar y las preparaciones permanentes se montaron con goma de resina de marañón (MGRC). El índice de refracción del MGRC fue de 1,343 y la viscosidad del medio sin aceite mineral fue de 1143,7 ± 267,4 mPas.sy con aceite mineral fue de 939,9 ± 103,7 mPas.s. Las preparaciones montadas con MGRC, independientemente del tipo de tinción utilizada, presentaron imágenes nítidas y morfología de las estructuras conservadas. El MGRC demostró ser una alternativa satisfactoria a los medios comerciales de montaje, siendo económicamente viable y generando beneficios como la bioseguridad personal al no ser tóxico, además de no dañar el medio ambiente.Neste trabalho, objetivou-se utilizar exsudato resinoso de uma planta típica do Nordeste brasileiro, a goma de cajueiro (Anacardium occidentale L.) como alternativa de meio de montagem na técnica histológica, economicamente viável e ambientalmente saudável. O índice de refração do MGRC foi 1,343 e a viscosidade do meio sem óleo mineral foi 1143,7 ± 267,4 mPas.s e com o óleo mineral foi 939,9 ± 103,7 mPas.s. As preparações montadas com MGRC independentemente do tipo de coloração utilizada, apresentaram-se com imagens nítidas e morfologia das estruturas conservadas.
