OPTIMIZATION OF GREEN PROPOLIS EXTRACTION BY ANTIFUNGAL ACTIVITY AND INCORPORATION IN CHITOSAN BIOFILM

Resumo: | The Brazilian propolis market has expanded greatly, leading the processing sector to pursue the best way to produce extracts with biological activities. The purpose of this research was to optimize the green propolis extraction by the antifungal activity and perform incorporation into a chitosan biofilm. The extracts were prepared using ethanol, acetone, ethyl acetate and isopropanol according to the three designs. The minimum inhibitory concentration (MIC) and the minimum fungicidal concentration (MFC) were determined against Candida albicans, Candida tropicalis e Candida glabrata. From the MIC, the response surface analysis was carry out to find the optimal conditions for the extraction for each design. The optimized extracts were incorporate into chitosan biofilm that were subsequently evaluate for thickness, water vapor permeation, water solubility and anti-Candida activity. It was establish that all extracts tested showed antifungal activity against the three Candida species. The extract obtained with acetone/ethanol stood out because it presented the lowest values of MIC and MFC (0.76 – 2.50 mg/mL). It was observe that the optimal extraction points of each design were the blend of acetone/isopropanol, acetone/ethanol and ethyl acetate/isopropanol yet the biofilms incorporated with these extracts showed few differences in thickness, vapor permeability and water solubility in relation to the control design. The biofilm obtained by acetone/isopropanol displayed the higher activity anti-Candida. This results show the importance of optimizing propolis processes for the application and product development. |

O mercado de própolis brasileiro sofreu grande expansão, levando o setor de beneficiamento a buscar a melhor forma de produzir extratos com atividades biológicas. O objetivo deste trabalho foi otimizar a extração de própolis verde pela atividade antifúngica e realizar a incorporação em um biofilme de quitosana. Os extratos foram preparados utilizando etanol, acetona, acetato de etila e isopropanol, de acordo com três planejamentos, e a concentração inibitória mínima (CIM) e a concentração fungicida mínima (CFM) foram determinadas contra Candida albicans, Candida tropicalis e Candida glabrata. A partir da CIM, realizou-se a análise de superfície de resposta, para encontrar as condições ótimas de extração para cada planejamento. Os extratos otimizados foram incorporados em biofilmes de quitosana que, posteriormente, foram avaliados quanto a espessura, permeação de vapor de água, solubilidade em água e atividade anti-Candida. Verificou-se que todos os extratos testados apresentaram atividade antifúngica contra as três espécies de Candida. Destacou-se o extrato obtido com acetona/etanol que apresentou os menores valores de CIM e CFM (0,76 - 2,50 mg/mL). Observou-se que os pontos ótimos de extração de cada planejamento foram as misturas de: acetona/isopropanol, acetona/etanol e acetato de etila/isopropanol e os biofilmes incorporados com estes extratos mostraram poucas diferenças de espessura, permeabilidade de vapor e solubilidade em água em relação ao controle. O biofilme contendo extrato obtido por acetona/isopropanol foi o que demonstrou maior atividade anti-Candida.
