Evaluation of different solvents combined with the high pressure technique for extraction of total phenolic compounds and antioxidant activity of yerba mate (ilex paraguariensis)

Resumo: It is increasingly common to use plants or derivatives to reformulate, replace or create food products, with the aim of adding nutritional values and satisfying the consumer who is increasingly demanding healthier foods. Yerba mate (Ilex paraguariensis) leaves are known for their antioxidant and antimicrobial potential, as well as improving glucose tolerance and anti-inflammatory effects. The objective was to analyze different solvents with the high pressure technique in order to obtain yerba mate extracts with a greater amount of total phenolic compounds and antioxidant activity. Ten treatments were performed, five subjected to high pressure extraction (150 MPa / 30 minutes) using as solvents: ethanol, H2O with ethanol, methanol, glycerin and lactic acid, and five not subjected to the technique. Regarding the solvent, the best results were for H2O + glycerin (50%). It was also observed that the highest content of phenolic compounds was in the extraction of yerba mate with pressure of 600.42 mg EAG/L, the same occurred with the antioxidant activity, and in the ABTS method the value was 619.70 TEAC /g without pressure and was increased to 702.53 TEAC/g with the use of the technique, for DPPH comparing before 954.37 IC50 (mg/mL) and after high pressure 1044.96 IC50 (mg/mL). It is concluded that high pressure extraction was essential to assist in the extraction of greater amounts of antioxidants using a biodegradable solvent.

Las hojas de yerba mate (Ilex paraguariensis) son conocidas por su potencial antioxidante y antimicrobiano, además de mejorar la tolerancia a la glucosa y sus efectos antiinflamatorios. El objetivo fue analizar diversos solventes con la técnica de alta presión para obtener extractos de yerba mate con mayor cantidad de compuestos fenólicos totales y actividad antioxidante. Se realizaron diez tratamientos, cinco sometidos a extracción a alta presión (150 MPa/30 minutos) utilizando como solventes: etanol, H2O con etanol, metanol, glicerina y ácido láctico, y cinco no sometidos a la técnica. En cuanto al solvente, los mejores resultados fueron para H2O + glicerina (50%). También se observó que el mayor contenido de compuestos fenólicos se presentó en la extracción de yerba mate con presión de 600,42 mg EAG/L, lo mismo ocurrió con la actividad antioxidante, y en el método ABTS el valor fue de 619,70 TEAC/g sin presión y se incrementó a 702,53 TEAC/g con el uso de la técnica, para DPPH comparando antes 954,37 IC50 (mg/mL) y después de alta presión 1044,96 IC50 (mg/mL). Se concluye que la extracción a alta presión fue fundamental para ayudar en la extracción de mayores cantidades de antioxidantes usando un solvente biodegradable.É cada vez mais habitual o uso de plantas ou derivados para reformular, substituir ou criar produtos alimentícios, com o objetivo de agregar valores nutricionais e satisfazer o consumidor que está cada vez mais exigindo por alimentos mais saudáveis. As folhas de erva mate (Ilex paraguariensis) são conhecidas por seu potencial antioxidante e antimicrobiano, além de tem efeito na melhoria da tolerância à glicose e efeitos anti-inflamatórios. O objetivo foi analisar variados solventes junto a técnica de alta pressão visando a obtenção de extratos de erva-mate com maior quantidade de compostos fenólicos totais e atividade antioxidante. Foram realizados dez tratamentos, cinco submetidos a extração por alta pressão (150 MPa / 30 minutos) utilizando como solventes: etanol, H2O com etanol, metanol, glicerina e ácido lático, e cinco não sujeitos a técnica. Em relação ao solvente os melhores resultados foram para H2O + glicerina (50%). Observou-se ainda que o maior teor de compostos fenólicos foi na extração de erva-mate com pressão 600,42 mg EAG/L, o mesmo ocorreu com a atividade antioxidante, sendo que no método de ABTS o valor foi de 619,70 TEAC/g sem pressão e foi aumentado para 702,53 TEAC/g com o uso da técnica, para DPPH comparando-se antes 954,37 IC50 (mg/mL) e depois da alta pressão 1044,96 IC50 (mg/mL).
